Den legendariske lejemorder John Wick har trukket sig tilbage, men da gangsteren Santino D’Antonia beder ham om at udføre et job, tvinges Wick til at efterleve den ed, han engang beseglede ved blod. Jobbet fører Wick til Rom, hvor han får hjælp af gamle bekendte, men da han opdager, at nogen har udlovet en dusør for at dræbe ham, ved han ikke, hvem han kan stole på.
